About TNBC Workbench

The overarching objective of this project is to reverse the course of the TNBC time machine. Key to this goal is the precise delineation of the mechanisms that lead to longitudinal progression of TNBC using innovative experimental approaches, multi-omics strategies and data analytics. This will lead to contextually precise markers, longitudinal resolution of mechanisms, deciphering of novel pathways and development of robust models of TNBC. Our combined expertise from City of Hope (CoH) and the University of California San Diego (UCSD) are:

• Availability of a unique and diverse CoH cohort of TNBC patients with linked and annotated biopsy tissue and patient-derived cell resources (e.g., patient derived organoids, tumor associated fibroblasts, adipose stromal cells, and normal cell strains derived from women at highest risk for TNBC)
• In vivo phenotyping capabilities (CoH).
• Complex omics characterization including scRNA/scATAC, snRNA, genotyping, and targeted spatial transcriptomics (UCSD).
• Innovative systems biology approaches to identify markers, mechanisms, pathways, and models (UCSD).
• Characterization of the longitudinal immune response and immune environment (CoH, UCSD).
• Longitudinal analysis to yield causal models (UCSD).
• Perturbation assays to validate hypothesis combined with in vitro therapeutic screening (UCSD, CoH)
• Follow-up with alternate cohorts (CoH).
